VZV infected cells are harvested and virus is extracted by alkaline treatment and sonication. The preparation is purified by ultracentrifugation through a sucrose cushion. The resulting antigen consists of a high concentration of virus and viral components, it may contain residual host cell material. The antigen is suspended in 100 mM glycine buffer, pH 9.5.
Physical inactivation. Inactivity is confirmed by attempted growth under original culture conditions. Since no procedure can guarantee absolute sterility, the reagent should be handled with appropriate safety precautions.
Protein Concentration: Lot specific, specified in mg/mL. Determined by Bradford assay (BioRad) using BSA as standard.
