Recombinant toxoplasma gondii Gra8 protein is expressed in E. coli as a monomeric, non-glycosylated polypeptide chain consisting of 149 amino acids with a molecular mass of 15800 Da. The product represents a central fragment of the native protein. It carries an N-terminal fusion of 6 Histidines.

The integrity of the plasmid is controlled by restriction analysis and DNA sequencing. The expression product is characterized by N-terminal protein sequencing, SDS-PAGE (app. m.w.) and immunodetection of the Western blotted protein. In addition, correct reactivities compared to previous antigen lots with defined human and animal sera (negative, low, medium and high positive) are mandatory.
