Uniprot ID Q8N6F7
Alternative Names Germinal center B-cell-expressed transcript 2 protein, Germinal center-associated lymphoma protein, hGAL
Gene Name GCSAM
Sequence Location Cytoplasm. Cell membrane. Note=It relocalizes from the cytoplasm to podosome-like structures upon cell treatment with IL6.
Biological Function Involved in the negative regulation of lymphocyte motility. It mediates the migration-inhibitory effects of IL6. Serves as a positive regulator of the RhoA signaling pathway. Enhancement of RhoA activation results in inhibition of lymphocyte and lymphoma cell motility by activation of its downstream effector ROCK. Is a regulator of B-cell receptor signaling, that acts through SYK kinase activation. {PubMed:17823310, PubMed:20844236, PubMed:23299888}.
Research Areas Cancer research
Background The human germinal-center-associated lymphoma (HGAL) gene was originally identified from an expressed sequence tag that was associated with improved survival in patients with diffuse large B-cell lymphoma (DLBCL). Cloning and characterization of HGAL indicated that it is induced by IL-4. HGAL is a cytoplasmic protein and has been demonstrated in follicular lymphoma, Burkitt lymphoma, lymphocyte-predominant Hodgkin lymphoma, and a subset of DLBCL. HGAL expression is an important predictor of overall survival in patients with germinal center- derived tumors. HGAL protein is also found in germinal centers of normal tonsils and lymph nodes.

Description Immunohistochemistry: use at 1-10ug/ml on paraffin-embedded tissues with heat-induced antigen retrieval in citrate buffer (10mM, pH 6.0, 10 minutes).
Description HGAL mRNA and protein expression in lysates of indicated cell lines. Actin served as a loading control.
Handling
Storage This antibody is stable for at least one (1) year at -20°C. Avoid multiple freeze-thaw cycles.
Dilution Instructions Dilute in PBS or medium that is identical to that used in the assay system.
Application Instructions
