Antibody Purification
QED will purify antibody from serum, ascites fluid, or
culture medium using a variety of purification procedures. These procedures
include ammonium sulfate or PEG precipitation, Protein-G chromatography,
and affinity chromatography.
Ammonium sulfate precipitation is a first
step in separating antibodies from other serum or ascites proteins. It
is useful for preparing antibodies for further purification by ion exchange
chromatography, or when you do not need a highly purified antibody preparation.
PEG precipitation is used to purify mouse IgM and IgG3
antibodies since they bind poorly to Protein-A and Protein-G.

Protein-G
chromatography yields pure antibody from serum, ascites
fluid, or culture medium without an initial preparatory step such as ammonium
sulfate precipitation. Purification on these media is rapid and can be
applied to small and large sample volumes.

Affinity chromatography
is used with polyclonal antiserum in which specific antibody comprises
only 5-10% of the total immunoglobulin. Affinity chromatography selectively
purifies the specific antibody of interest from the remaining antibodies
in the antiserum.

Antibody-Reporter Conjugations
Antibody-Reporter Conjugations QED is experienced in enzyme,
biotin, and fluorophore conjugation techniques. By conjugating these labels
to your antibody, you can create a primary immunoreagent. Directly labeling
your primary antibody allows you to eliminate steps in your assays, saving
time and money. QED can assist you in determining which reporter molecule
is best for your application.

Hapten-Carrier Conjugations
Small haptens, or poorly reactive antigens, are often made
more antigenic when conjugated to carrier proteins, such as keyhole limpet
hemocyanin. QED will assist you in determining whether conjugation is
appropriate for your antigen, and which carrier protein is best for your
application

ELISA Development and Testing
ELISAs (Enzyme Linked Immunosorbent Assays) are used to
measure the specific binding of an antibody to an antigen. This is a rapid
and sensitive method for evaluating large numbers of monoclonal or polyclonal
antibodies at low cost. QED scientists have years of experience developing
ELISAs for our clients and for our own in-house antibodies and we are
pleased to share that experience with you. Once an ELISA has been developed
for you, we will transfer the ELISA protocol to you and, if you wish,
apply that ELISA to testing of your antibody samples.
